⑷ Basic requirements of the procurement :
A.Field-emission scanning electron microscope (FE-SEM) : 1 unit.
a It should be equipped high performance apparatuses for electron beam, lens, and detector that are capable for high resolution observation of biological samples.
b It should be capable to observe ultra-thin sections by transmission methods and to acquire high-resolution images.
c It should be capable of high-resolution observation in a low-vacuum environment that allows biological samples to be observed as close to the natural state as possible.
B.Energy dispersive X-ray spectroscopy system : 1 unit.
a Enables elemental analysis and mapping at the arbitrary sites in samples.
C.Light/electron-correlated observation unit : 1 set.
a It should be possible to make correlate images in both directions between the FE-SEM and the inverted fluorescent microscope that owned in the Joint Faculty of Veterinary Medicine, Yamaguchi University.
b To be able to adjust the coordinate axes of the two instruments and to enable superimposition of observation images.
D.3D image construction/analysis system : 1 set.
a A microtome that capable of preparing serial sections from resin-embedded samples.
b A sample stage that enables efficient observation of the above serial sections in the FE-SEM.
c Software that enables creation of 3D images from the above observation images from FE-SEM, analysis of arbitrary structures of the images, and application of images and movies for presentation slides.
