⑷ Basic requirements of the procurement : Requirements for the complete set of the transmission electron microscope for cryo-EM are described below.
A A transmission electron microscope for cryo-EM equipped with a cold field emission gun (FEG)
a It should be equipped with a cold FEG, which produces a highly coherent electron beam. The energy resolution is supposed to be less than 0.4 eV without using a monochromator.
b The accelerating voltage should be higher than 300 kV.
c The automated specimen exchange system should be equipped. It must be able to contain up to 12 grids at the same time.
d The system should contain a direct electron detector (DED). The physical pixel size of the DED should be less than 5 μm2 and it has 24 megapixels (5,760x4,092) field of view. The readout speed of the DED should be 1,500 full frames per second. It contains the GPU-based motion correction feature.
B A vacuum evaporator This is needed to prepare amorphous carbon layer for grid preparation.
a The reachable vacuum degree should be higher than 1x10-4Pa.
b A cold trap should be equipped.
C A glow discharger This is needed to hydrophilize the surface of the grid for grid preparation.
a The size of the electrostatic shield electrode should be larger than 50 mm in its diameter.
D An automated grid plunging devise This is needed to perform plunge-freezing for vitrification of specimens.
a The temperature of the specimen chamber should be controllable at the range from 4℃ to 60℃.
b The container should keep 1 ℓ liquid nitrogen for one hour.
